MOLECULAR RESOLUTION FACILITY (MRF)

The MRF is equipped for resolving complex extracts and synthetic samples and identification of components using high performance liquid chromatography. Our bioelectrochemistry laboratory enables analysis of redox active proteins, screening of (potential) drugs and determination of properties and activities.

MRF is equipped with the latest Dionex HPLC (analytical and preparative) systems driven by Chromeleon Software, providing state-of-the-art chromatography capabilities in separation, purification as well as qualitative and quantitative HPLC services.

Instrumentation

System 1: Dionex Analytical HPLC with Alltech 2000ES ELSD
The Dionex Analytical HPLC system comprises the P680 quaternary gradient pump, an ASI-100 automated sample injector capable of injections from 1-250 mL, an on-line solvent degasser and a thermostatted column compartment, which contribute to reproducibility. The system is equipped with two detectors, the workhorse PDA-100 photodiode array detector and an Alltech 2000ES Evaporative Light Scattering Detector (ELSD). The PDA-100 is a high-resolution detector with two light sources, a deuterium lamp for UV wavelengths, and a tungsten lamp for visible wavelengths, which together provide a broad (190-800 nm) spectral range. The 2000ES ELSD is useful for the analysis of analytes, which lack UV chromophores.

System 2: Dionex Preparative HPLC with Foxy Jr Fraction Collector
The Dionex Preparative HPLC system, in addition to the same basic components as the analytical system, employs a UVD 340U PDA detector and a Foxy Jr fraction collector. The system is capable of fully automated fraction collection/purification (sample tracking) with collection based on peak recognition and/or time. The preparative scale ASI-100 sample injector is capable of injections up to 2.5 mL. The preparative HPLC pump is capable of delivering eluent at a rate of 0.2-100 mL/min whilst operating isocratically. The sample carousel can be configured to house 1.2 mL vials (192), 2 mL vials (117), or 4 mL (63) vials. The UVD 340U utilises a selection of flow cells allowing for a range of applications from microbore LC-MS to preparative HPLC.

Example of Fraction Collection and Sample Tracking
Fraction collection can be undertaken on the Foxy Jr Fraction Collector using methods including microtitre plate, small fractions for possible MS analysis, test tubes as well as collection into small and large bottles via the use of funnels. Sample tracking is possible using the Chromeleon Software fraction collection software on the preparative HPLC (System 2).

Fraction collection can be programmed to collect based on peak recognition at different threshold values. Collection of regions between peaks can also be programmed.

Semi-preparative HPLC columns: Reverse phase (C18 Gemini, Phenomenex and Alltech 250 x 10 mm, 5 µ and Normal Phase Silica Alltech 250 x 10, 5 µ)

Gemini 5µ C18 column (250 x 10mm) using a
water/acetonitrile gradient at 4.5mL/min. Collection based
